I used the same picture you did, what did you do to it? I just used the magic wand tool (with AA on) to cut out the white part and the shadow behind the emblem. I then took the shape of the emblem, added a layer behind it, filled it with black, set the opacity to 75% and feathered to edges to give it a soft shadow that will be transparent in the avatar. After that I resized it and voila!

Well the pattern appears like that because MS Paint is too simple to understand the complexity of a transparent background, so when I opened the file up using MS Paint for editing, I got the picture that I attached earlier except it was using inverted colors.
Try this test: Invert the colors of my inverted picture and open it up using a picture viewing program other than MS Paint. It should turn out the way you planned.

Here is a good tip for making transparent backgrounds, Carcrazy.
First start your signature (120x550 px) with a transparent setting. Don't do anything to it yet.
Next get a car image for your signature (make sure its a clear image!) and use the polygonal lasso to tediously select the car from the image.
After the car is selected and placed into your signature, resize it to a size you like then ctrl + click on the layer of your car. Go to Select -> Feather and feather it by 2 or 3 px.
Invert your selection by pressing ctrl + i and then press delete on your keyboard once or twice depending on how much you want to soften the edges of the car.
Next put in your cool font and add some blending effects to it (the little black button on the layers palate or double click the font layer) then make a new layer and press ctrl + a to select all and go to Edit -> stroke and set it for black 1px inside and click ok. Then you have a nice simple transparent signature. Make sure though that you go to File -> Save for Web and set it to PNG - 24 with interlaced on.
